PGC-1α (peroxisome proliferator-activated receptor gamma coactivator 1-alpha) is a critical transcription coactivator serving as a primary regulator of energy metabolism and mitochondrial biogenesis. Highly expressed in tissues where oxidative metabolism is active—such as brown adipose tissue, heart, and skeletal muscle—this protein regulates glucose and fatty acid metabolism by interacting with nuclear receptor family members including PPAR-α, PPAR-γ, and PPAR-β/δ, as well as transcription factors such as MEF-2 and SIRT1. Increased neuromuscular stimulation and contractions during exercise significantly induce PGC-1α gene expression. PGC-1α stimulates the conversion of skeletal muscle fiber types from glycolytic (type II) muscle fibers to fatigue-resistant oxidative (type I) muscle fibers; it elevates myoglobin and myosin heavy chain expression, mitochondrial density and respiratory capacity, glycogen storage, and fatty acid oxidation capacity. Research studies in the literature demonstrate that high-intensity interval training (HIIT), endurance exercises, exercises under hypoxic conditions, electrical stimulation, and alpha-lipoic acid supplementation significantly increase cellular PGC-1α levels. Consequently, PGC-1α plays a key role in meeting energy demands, oxygen transport, enhancing fatigue resistance, and supporting fat burning, thereby contributing to performance enhancement and biochemical adaptation to exercise in both athletes and sedentary individuals.
