Özet

Patoloji, hastalıkların vücut yapısı ve işlevindeki etkilerini inceleyen, klinik öncesi ve klinik bilimler arasında köprü kuran temel bir tıp disiplinidir. Tarihsel kökleri antik çağa uzanan bu bilim dalı, 19. yüzyılda mikroskobinin gelişmesiyle modern kimliğine kavuşmuştur. Günümüzde genel ve sistemik patoloji olarak iki ana gruba ayrılan alan; morfolojik (histopatoloji, sitopatoloji, hematoloji) ve morfolojik olmayan (klinik biyokimya, mikrobiyoloji, immünoloji vb.) dalları barındırır. Histopatolojide dokular, otolizi önlemek amacıyla formaldehit gibi sıvılarla fikse edilir, ardından dehidrasyon, şeffaflandırma ve parafin infiltrasyonu aşamalarından geçirilerek bloklanır. Mikrotomla alınan ince kesitler, rutin olarak hematoksilen-eosin yöntemiyle boyanarak ışık mikroskobunda incelenebilir hale getirilir. Ameliyat sırasında hızlı tanı için kriyostat cihazıyla dondurma kesiti (frozen) tekniği de kullanılır. Sitopatolojide ise vücut yüzeylerinden dökülen veya iğne aspirasyonu ile alınan hücreler, alkol fiksasyonunun ardından PAP veya Giemsa gibi yöntemlerle boyanarak incelenir. Patoloji laboratuvarları; kayıt, makroskopi, doku işlem, mikroskopi, raporlama ve arşiv bölümlerinden oluşarak kesin tanı sürecini yürütür.

Pathology is a fundamental medical discipline that examines the effects of diseases on body structure and function, serving as a vital bridge between preclinical and clinical sciences. With historical roots extending back to antiquity, this field achieved its modern identity in the 19th century through the development of microscopy. Today, divided into two main categories as general and systemic pathology, the discipline encompasses morphological branches such as histopathology, cytopathology, and hematology, as well as non-morphological branches including clinical biochemistry, microbiology, and immunology. In histopathology, tissues are fixed with fluids like formaldehyde to prevent autolysis, and are then blocked by undergoing stages of dehydration, clearing, and paraffin infiltration. Thin sections obtained via microtome are routinely stained using the hematoxilyn-eosin method, making them examineable under a light microscope, while the frozen section technique with a cryostat device is utilized for rapid diagnosis during surgery. In cytopathology, cells shed from body surfaces or collected via fine-needle aspiration are examined by being stained with methods like PAP or Giemsa following alcohol fixation. Pathology laboratories execute the definitive diagnosis process by consisting of material registration, macroscopy, tissue processing, microscopy, reporting, and archive departments.
