The reproductive cycle in rats (estrus cycle) differs from humans, featuring a polyestrous structure with spontaneous ovulation that repeats every 4 to 5 days. The estrus cycle of female rats, whose gestation period lasts approximately 20 days, is examined in four main phases based on the cell typology they contain: proestrus (12 hours), estrus (9–15 hours), metestrus (21 hours), and diestrus (57 hours). While ovulation occurs between the end of proestrus and the beginning of estrus, the proportions of nucleated epithelial cells, cornified squamous epithelial cells, and leukocytes in the vaginal smear allow the determination of these stages. Significant histological changes occur in the vagina, uterus, and ovaries throughout the cycle; the uterine epithelium, which is cuboidal or prismatic in proestrus, degenerates during estrus and metestrus, and transitions to a low columnar structure in diestrus. In the female reproductive system, estradiol, progesterone, and prolactin hormones play active roles alongside FSH and LH gonadotropins released from the pituitary gland via GnRH stimulation. LH, which peaks during proestrus, stimulates ovulation, while progesterone secreted from the corpus luteum formed after ovulation supports uterine thickening. While the uterus is maintained steadily by progesterone in metestrus, the decrease in progesterone and the increase in estradiol during diestrus restart the follicular development cycle.
