This text explains how to determine the stages of the estrous cycle (proestrus, estrus, metestrus, diestrus) in rats using vaginal smear methods. The classification of the stages is based on the presence, absence, or ratio of neutrophils, nucleated, and anucleated epithelial cells, as well as cell density and the arrangement of cells on the slide. The first step in the evaluation is assessing the presence of neutrophils; if neutrophils are a dominant feature, the stage is either metestrus or diestrus, whereas if they are rare or absent, the stage is either proestrus or estrus. In the proestrus stage, typically no neutrophils are observed, while uniform, small-sized, round, and deeply basophilic staining nucleated epithelial cells are distinguished in clusters or sheets. During the estrus phase, anucleated keratinized epithelial cells and free bacteria are predominantly seen, and neutrophils are generally absent. The metestrus stage is characterized by a combination and dominance of anucleated keratinized epithelial cells and small, darkly staining leukocytes (neutrophils). In the diestrus stage, a significant decrease occurs in the number of anucleated keratinized epithelial cells, and the number of neutrophils is generally much higher compared to epithelial cells. Furthermore, the presence of nucleated epithelial cells in diestrus differentiates this stage from metestrus.
