Farklı Vajinal Sitoloji Tekniklerinin Karşılaştırılması

Vajinal yayma örneklerinin boyanmasında Papanicolaou (Pap), Giemsa, Diff-Quick ve H&E gibi birçok yöntem kullanılmaktadır. Pap boyaması; nükleer detayların incelenebilmesi, sitoplazmik şeffaflık ve skuamöz epitelin hücresel farklılaşmalarının gösterilebilmesi gibi avantajlara sahiptir ve hücrelerin olgunluk düzeyine göre renk ayrımı sağlayarak proöstrus evrelerinin anlaşılmasını kolaylaştırır. Ancak fiksasyon gerektiren bu protokol 25-30 dakika sürer ve diğer boyalara göre daha maliyetlidir. Kristal Viyole, Metilen mavisi, Toluidin mavisi ve H&E yöntemlerinde ise hücrelerde evrelere göre renk değişikliği gözlenmez. Wright-Giemsa boyası, sitoplazmik detaylarla hücre-nükleer oranının tahmin edilmesini sağlar ve fiksasyon gerektirmez ancak keratinizasyonu vurgulayamaz. In the staining of vaginal smear samples, many methods such as Papanicolaou (Pap), Giemsa, Diff-Quick, and H&E are utilized. Pap staining offers advantages such as the examination of nuclear details, cytoplasmic transparency, and the demonstration of cellular differentiations of the squamous epithelium, facilitating the understanding of proestrus stages by providing color differentiation based on the maturity of the cells. However, this protocol, which requires fixation, takes 25-30 minutes and is more costly compared to other stains. Conversely, no color change is observed in cells according to different stages when using Crystal Violet, Methylene blue, Toluidin blue, and H&E methods. Wright-Giemsa stain allows for a better estimation of the cell-nuclear ratio with improved cytoplasmic details and does not require fixation, but it fails to emphasize keratinization. Toluidine blue and Methylene blue methods are advantageous techniques that do not require any special fixation, are simple, take less time, and clearly demonstrate structures like bacteria. Additionally, the direct examination of unstained vaginal cytology samples under the microscope is a fast, practical, time, and cost-advantageous method; however, the evaluation must be performed carefully by experienced personnel.
